　　1.用含10%小牛和的RPMI-1640培養(yǎng)液將K562細胞配成1&time105/ml濃度。用同樣培養(yǎng)液將MC配成1&time106/ml濃度。
　　2.取96孔圓底，每孔加0.1ml K562細胞懸液;每孔加適量MC使效靶比為1：1～5：1，每種處理3個復(fù)孔;3個對照孔只加效應(yīng)細胞;8個對照孔只加靶細胞;4個對照孔只加細胞培養(yǎng)液。每孔加20l Alamar Blue;每孔總量為0.2ml。
　　3.在37℃ 5% CO2的飽和水汽二氧化碳培養(yǎng)箱中培養(yǎng)24小時。
　　4.直接在熒光檢測儀上檢測熒光強度，激發(fā)波長530nm，發(fā)射波長590nm。各孔熒光強度值應(yīng)減去培養(yǎng)液對照熒光強度的平均值，各復(fù)孔熒光強度的平均值記為AF。按下式計算特異性殺傷百分比：
